Basic reporting

This is a well written review with helpful complementary figures.

Given vascular dysfunction, specifically large artery stiffening and endothelial dysfunction, are major antecedents to cardiovascular disease, this review would greatly benefit from discussion regarding the role of TFEB in these processes (there is literature to support this connection.

Basic reporting

The TFEB and cardiovascular disease review was already published in EBioMedicine in 2021 (https://www.thelancet.com/action/showPdf?pii=S2352-3964%2820%2930583-1). Some of the discussion in this manuscript is already clearly described in the previous EBioMEdicine paper. The authors need to add much information that is still not discussed yet in the previous paper.

Experimental design

Some of the previous studies mentioned by the authors are still in vitro studies. It’s better to add information in one summary of the role of TFEB in each CVD both in vitro and in vivo studies, so the readers will understand which role of TFEB still needs addition to vivo studies.

The conclusions are well stated.
