Another amyloid lyophilization question

Hi Ryan,

I have another question regarding amyloid lyophilization. Recently, we bought Labconco 2.5L, -84°C benchtop lyophilizer and I tried to lyophilize amyloids with it. I prepared amyloids with NH4OH method. Basically, I added 1ml of 1% NH4OH solution to 1mg of amyloid beta (1-42), I mixed it slightly by hand, to dissolve it. Then I sonicate solution at RT for 5 min, aliquote into 50ul aliquots and put them into -80 freezer for 30 min. After they were frozen, I put them into lyophilizer. Before sample insertion, lyophilizer temp was set to -84°C and vacuum was the highest possible one - 0.003mbar. I used highest vacuum because you said that you used the highest one that lyophilizer can provide. The problem is that after 24h sample is still in form of ice (basically like nothing happened, it looks the same as when I took it from freezer). You can see the sample on this photo - https://imgur.com/a/8aN9WSI. So, I don't know what's the problem. Maybe the pressure is too low and that's why drying is so slow? Any kind of advice/help is welcomed. Thank you in advance.
